Objective To determine whether gp130 associated molecule (GAM) interacts with Transducin Like Enhancer of split 1 (TLE1) molecules, and locate the domains involved in this interaction. Methods The cDNA fragments encoding various domains of GAM and TLE1 were amplified by PCR and cloned into the yeast duplicate hybrid vectors pGAD424 or pGBT9, yeast duplicate hybrid assay and immuno-co-precipitation assay were performed to verify the interaction between GAM and TLE1. Results The yeast duplicate hybrid vectors containing cDNAs of various domains of GAM and TLE1 were successfully constructed. Yeast duplicate hybrid assay and immuno-co-precipitation assay indicated that GAM and TLE1 interact by their homologous Q domain. Conclusion GAM and TLE1 interact by their homologous Q domain.
